The differential sucrose-dependent cariogenic traits of Streptococcus mutans in the presence of dietary carbohydrates

Abstract
Objectives: The aim of this study was to investigate the differential sucrose-dependent cariogenic traits of Streptococcus mutans in the presence of dietary carbohydrates.

Methods: S. mutans KCTC3065 were cultured in TYE medium containing 0.5% glucose at 37oC under 10% CO2 for 24 hours. The strain was divided into the following 5 groups in terms of the culture medium that was used: (1) 0.5% glucose containing basal culture TYE medium (BCM), (2) BCM plus 0.5% sucrose, (3) BCM plus 0.5% sucrose and fructose, (4) BCM plus 0.5% sucrose and maltose, (5) BCM plus 0.5% sucrose, fructose and maltose. After the cells were collected, growth, acid production synthesis of extracellular polysarcharides and related gene expression were evaluated.

Results: The growth and acid production of S. mutans in the presence of sucrose and fructose were delayed and decreased, but not in the presence of sucrose and maltose (p£¼0.05). The expression level of virulence-related genes associated with gtfB and gtfD were significantly difference under all conditions (p£¼0.05). Moreover, there were more extracellular polysaccharides due to gtfB and gtfD expression in the presence of sucrose, fructose, and maltose.

Conclusions: These results indicate that the growth and acid production of S. mutans were affected by the presence of sucrose, fructose and maltose, which also effectively increased extracellular polysarccharides synthesis by gtfB and gtfD expression. Thus, sucrose-dependent cariogenic traits of S. mutans can be increased.
